Next Thursday, October 12, the Sanger Core will begin accepting sequencing orders on Mondays, Tuesdays, and Thursday.
Open services
- High Throughput Sequencing (HTS)

Schedule and turnaround time
- We will begin processing new orders at 10am on Mondays, Tuesdays and Thursdays
- Expected turnaround time is 48 hours from the 10am Monday/Tuesday/or Thursday intake
- You will receive an email about your order at intake if we do not expect to be able to meet the 48-hour turnaround goal

- Dropoff submissions in the freezer outside the Sanger Core (KCBD 1230H)

Order and sample submission updates
- There are some updates to facilitate faster sample processing and data turnaround while limiting sample mix-ups.
- Complete, updated instructions for HTS and LTS are attached and I will bring your attention to some key changes below. Complete instructions will be updated on https://dnaorder.uchicago.edu/ by early next week
- High throughput order updates:
- HTS orders must contain at least 8 reactions submitted in strip tubes or 96 well plates.
- HTS orders with custom primers should be submitted as a premix of template (4.5 uL) and primer (3 uL at 4uM). Your submission to the core freezer should include 7.5 uL of premix in a distinct well for each ordered reaction. In the order template use the term “premix” in the primer column. If you would like your primer included in the filename of your returned sequence, include the primer name in the Sample Name column.
- HTS orders that include only core-supplied primers must include a minimum of 8 reactions per distinct primer. Your submission to the core freezer should include 4.5 uL of template* in a distinct well for each ordered reaction
- Low throughput order updates:
- LTS orders of fewer than 8 reactions may be submitted in individual 0.2mL tubes, 1.5 mL tubes, or strip tubes. Orders of 8+ reactions should be submitted in strip tubes.
- For LTS orders, submit 4.5 uL of template in a distinct well for each reaction. Submit custom primers (3uL at 4uM per reaction) in 1.5 mL tubes labeled with the primer name as entered in the order template.
- Sample labeling updates: Label tubes or plates by the well position (A1, B1, C1…12F, 12G, 12H), NOT the template name or primer. Strip tubes and plates only require that the first well of each be labeled. Please also label each 1.5mL tube and strip tube with your PI’s last name and your first and last initial.
I am excited to have a new staff member joining me on October 16 and I am optimistic about the future of the Sanger core. On that note, here are some future goals that we will be working towards:
- Accept new orders daily
- Re-instate genotyping and colony picking services
- 30-hour turnaround times
Finally, I want to thank you all again for your patience with me as I have struggled to respond to many emails and work through the backlog of samples. Those of you that I have been able to connect with have been incredibly gracious and understanding and I truly appreciate that. Please continue to bear with me through the first couple of weeks of reopening the core. I realize that I have made changes to a system that you are very familiar and accustomed too, and I hope you find these reasonable to the end of returning quality data to you faster. I’m eager to move forward and to provide you reliable Sanger sequencing services.
