10x Genomics
The 10x genomics Chromium X combines flow cytometry, microfluidic dropletization, and unique molecular identifiers (UMIs) to produce single-cell resolved sequencing libraries. A cell suspension is partitioned into droplets, and nucleic acid molecules are bound and tagged by specialized beads coated in oligonucleotides. These oligonucleotides contain sufficient UMIs to distinguish up to 20k individual cells. The ligated fragments are then prepared as illumina sequencing libraries. Submission Guidelines:
10x genomics projects require an approach tailored to the clients needs. We use the following general procedure for facilitating 10x experiments with our clients.

The client selects a 10x workflow that meets their research objectives.

The client submits a request for 10x sequencing (link to form)

The client prepares single-cell suspensions (Cytometry and Antibody Technology – an OSRF Core Facility at UChicago).

Client brings single-cell suspensions to the facility for a ‘dry run’. The ‘dry run’ is a simple validation experiment to ensure cell concentration and viability are sufficient for a successful 10x experiment.

The client will work together with our facility to schedule a full 10x experiment. Our facility will order the reagents needed to complete the workflow chosen by our client.

The client will deliver the cell suspension(s) needed for the full experiment. Cells are partitioned and used to make single cell resolved libraries.

Libraries are processed and sequenced in our facility.
Cell Suspensions must meet certain criteria for successful 10x experiments. Suspensions with significant numbers of dead cells or cellular debris may impact recovery rate. Below are the manufacturer recommendations for 10x cell suspensions.
