Methylation Sequencing
We offer methylation sequencing using either the New England Bio Labs (NEB) enzymatic methyl-seq, and Enzymatic 5hmC-seq kits. For both methods undecorated cytosines are deaminated to uracil, while decorated cytosines are not. Subsequent sequencing of the whole genome library in species with known sequence allows for information about methylation to be collected. For the standard methyl-seq kit, both 5mC and 5hmC are protected from conversion, while the 5hmC kit only protects 5hmC decorated cytosines from deamination. For methylation sequencing clients should submit genomic DNA at concentrations at or above 100 ng/µl. Necessary reagents are stocked by the facility.
