Services

Overview:  The HIM Facility provides laboratory support to UCCCC investigators who conduct novel immunotherapy clinical trials. The services include multidimensional biobanking, standardized immune monitoring assays, and novel single cell and/or tissue-based assays.

 

Services
  • Isolate the bank PBMCs, serum, and plasma from whole blood
  • Various tissue banking and stool banking
  • Patient tissue specimen dissociation and single cell library preparation for sequencing
  • DNA, RNA isolation from whole blood, fresh frozen and/or archival FFPE tissue specimen, various body fluids, and fecal material for downstream genomic and sequencing applications
  • Multiplex IF in FFPE tissue sections (up to 6 protein markers plus DAPI)
  • Whole slide scan using Vectra Polaris Imaging System and data analysis using Inform software
  • In situ mRNA and protein marker analysis in FFPE tissue sections using RNAscope plus IF (up to 2 RNA markers and 2 protein markers plus DAPI)
  • Cell culture including pan T cell expansion and antigen-specific CD8+ T cells expansion
  • ELISPOT assay
  • Flow cytometric staining and/or sorting for hematopoietic subsets, activation markers and intracellular molecules in peripheral blood, including Treg quantitative analysis
  • ELISAs for specific factors in serum or plasma and culture supernatants
  • Real-time RT-PCR analysis of immune-related genes, tumor antigens, or other relevant transcripts
  • Gene expression analysis in archival FFPE tissues using Nanostring technology
  • Single cell secretome analysis using Isolight system
